摘 要:[目的]分析碳离子辐照对甜高粱幼苗叶片中保护酶同工酶的影响。[方法]以甜高粱品种KFJT-CK为对照,以经碳离子辐照选育的早熟突变株KFJT-1为试材,采用垂直板聚丙烯酰胺凝胶电泳法(PAGE)对幼苗叶片中的过氧化物酶(POD)、酯酶(EST)和超氧化物歧化酶(SOD)同工酶谱进行了分析。[结果]突变后代KFJT-1与对照KFJT-CK相比,POD同工酶酶谱基本相同,说明碳离子辐照并未使甜高粱POD同工酶的表达发生明显的变化;SOD同工酶酶谱的条带数相同,只是条带的颜色深浅有所差异,说明碳离子辐照仅使SOD同工酶的表达量有所差异;EST同工酶增加了1条新谱带,说明碳离子束辐照使甜高粱EST的表达发生了变异。[结论]碳离子辐照对甜高粱幼苗叶片中POD的影响很小,对SOD有较微弱的作用,而对EST的同工酶谱有很大的影响。[ Objcetive ] Analyze the effects of carbon ion beam irradiation on the expression of protective enzymes isozymes in sweet sorghum seedling leaves. [ Method ] Using KFJT-CK and early-maturity mutant KFJT-1 , cultivated from KFJT-CK that treated by carbon ion beam ,as experiment materials , the isozymes of peroxidase (POD) , esterase (EST) and superoxide dismutase (SOD) were analyzed through polyacrylamide gel electrophoresis( PAGE ). [ Result ] Comparing KFJT-1 with KFJT-CK, the POD isozyme electrophoretogram was much the same which indicted that the expression of POD was not changed apparently by carbon ion beam irradiation. As for SOD, . the bands number were the same, but the color of some bands varied, which showed that the expression of SOD isozyme was slightly affected . The electrophoretogram of EST isozymes generated a new band , suggesting that the expression of EST in sweet sorghum was affected largely by carbon ion beam. [ Conclusion] carbon ion beam irradiation had no much effect on the expression POD isozymes, slight effect on the SOD expression and significant variation on the expression of EST isozymes.
