摘 要:[目的]探讨生黄芩饮片黄酮类化合物的纤维素酶辅助提取优化工艺条件。[方法]采用系列单因素和正交试验设计,通过高效液相分析以黄酮苷转化成的苷元含量为考核指标,对影响辅助提取的因素进行了研究,并与传统水煎煮法比较。[结果]生黄芩黄酮苷元的最佳纤维素酶辅助提取工艺为液固比10∶1,酶浓度25 U/g(药材),pH值5.0,温度50℃,提取时间7 h。最佳提取条件下,黄酮苷元黄芩素和汉黄芩素的提取率分别为175.04、28.84μmol/g(药材),分别为传统水煎煮法的6.7倍和4.8倍。[结论]纤维素酶辅助提取法提取黄芩素和汉黄芩素等转化率高,易于实现规模化生产。[ Objective ] The research aimed to study the optimum extraction technique of flavonoid aglycons from skullcap root by cellulaseassisted extraction method. [ Method] The conditions for the extraction by cellulase-assisted extraction method were studied through several single factor tests and the orthogonal experimental design, as an evaluation index of the content of baiealein and wogonin in skullcap root by HPLC method. The cellulase-assisted extraction method was compared with the water-deeocting method. [ Result] The optimized conditions of cellulase-assisted extraction are as follows: liquid-sold ratio is 10:1, the concentration of cellulase is 25 U/g herbals, pH value is 5.0, extrac- tion temperature is 50 T: , and extraction time is 7.0 hours. Under the optimal conditions, the extraction rate of baiealein and wogonin reached 175.04 and 28.84 μmol/g herbals respectively, being 6.7 times and 4.8 times as that of the conventional water-decoeting method. [ Conclusion ] Cellulase-assisted extraction method for the flavonoid aglyeons in skullcap root was efficient, appropriate and feasible.
