摘 要:目的从血红素氧合酶-1-CO系统的角度探讨中药复方四逆汤抗心肌缺血再灌注损伤(IRI)的机制。方法48只雄性昆明小鼠随机分为正常对照组、缺血再灌注模型组、四逆汤处理组,垂体后叶素联合硝酸甘油复制心肌缺血再灌注模型,采用双波长分光光度法间接测定血浆及心肌组织一氧化碳(CO)浓度、Western blot分析心肌组织血红素氧合酶-1(HO-1)蛋白的表达。结果IR模型组血浆和心肌组织CO浓度明显高于对照组,而四逆汤处理组较模型组又明显增加。Western blot显示模型组、四逆汤组在造模30min后,出现少量HO-1蛋白表达,2~4h达到高峰,至24h仍有少量表达;图像扫描灰度值比较,四逆汤组HO-1蛋白相对表达量在不同的时间点位均高于模型组,约为模型组的1~2.5倍。结论四逆汤能增加HO-1蛋白的表达,促进血红素分解,升高内源性CO浓度,调节血红素氧合酶-1-CO系统,发挥全面的细胞保护作用可能是四逆汤抗心肌IRI的重要机制之一。Objective: To discuss the mechanism of Sini Decoction on heme oxygenase-1-carbon oxide (HO-1-CO) pathway in cardiac ischemia reperfusion injury. (IRI) rats. Methods: 48 male mice were randomly divided into the control group, the IRI model group, the Sini Decoction group. The IR models were established by hypophysin combined with nitroglycerin. The CO concentrations in plasma and myocardial tissue were detected by double wavelength spectrophotometer. The HO-1 expression in cardiac tissue was investigated by the western blotting method. Results: With CO concentration in plasma and myocardial tissue, the IR model group was higher than control group. However, it was Dower than the Sini Decoction group. After modeling 30min, a few of HO-1 proteins were expressed in the model group and the Sini Decoction group. In 2 to 4 hours, these proteins expressed the top concentration. In the 24th hour,its expression was still occurred. Compared with the gray valued of image scanning, the HO-1 protein expressions of Sini Decoction group were higher than those of model group in different time point. It was approximate 1 to 2.5 times of model group. Conclusion: Sini Decoction could not increase the levels of HO-1 expression in myocardial tissue and promote hemoglobin decomposition, but also raise CO concentration in plasma and regulate the HO-1-CO system. This would be one of the most important mechanisms of Sini Decoction against cardiac iscbemia reperfusion injury.
