摘 要:目的观察化合物康普瑞丁磷酸二钠(CA4P)对人宫颈癌Hela细胞的体外抑制作用及对裸鼠移植瘤的体内抗癌作用。方法用MTT法研究CA4P对Hela细胞增殖的影响,将Hela细胞移植于裸鼠皮下,观察肿瘤的生长状况,并计算抑瘤率和脏器系数。结果CA4P0.5~64μg·mL-1时,对Hela细胞24、48h的抑制率分别为15%~75%、18%~80%,IC50分别为0.336、0.207μg·mL-1;对裸鼠Hela细胞移植瘤的抑制率为30%~50%,且呈量效关系。结论CA4P对人宫颈癌Hela细胞的增殖具有抑制作用,对裸鼠移植瘤的体内抑制作用明显。OBJECTIVE To observe the inhibitory effect of Combretaflation A4 prodrug( CA4P )on human cervical carcinoma Hela cell in vitro and in nude mice. METHODS The inhibitory effect of CA4P human cervical carcinoma Hela cell was investigated by MTT analysis. Hela cells were inoculated subcutaneously into nude mice. Volumes of tumor were measured and growth of tumor was observed and the inhibitory rate of tumor growth was computed. RESULTS After treated with CA4P for 24 or 48 hours, MTT assay showed the dose range from 0.5μg·mL^-1 to 64μg·mL^-1 had the inhibitory ratio range from 15% to 75% and from 18% to 80% respectively. The ICs0 was 0. 336μg·mL^-1 and 0.207 μg·mL^-1 , respectively. The inhibitory rate of tumor growth in nude mice carried turnout was 30% to 50% , and it was related with the dose. CONCLUSION CA4P could inhibit the growth of human cervical carcinoma Hela ceil in vitro and in vivo.
