摘  要:目的建立测定芪桂消癥颗粒中黄芪甲苷含量的方法。方法采用HPLC-ELSD法,色谱柱为Diamonsil C18柱(150mm×4.6mm,5.0μm),流动相为乙腈-水(32:68),流速为1.0mL·min-1,柱温为35℃;检测器漂移管温度为80℃,氮气压力为45psi。结果黄芪甲苷7.72~23.16μg与峰面积的线性关系良好(r=0.9993),平均回收率为97.49%,RSD=2.08%(n=6)。结论所建方法简便、准确、重复性好,可有效地控制芪桂消癥颗粒的质量。OBJECTIVE To establish a method to determine the content of astragaloside IV in Qigui Xiaozheng granule. METHODS HPLC -ELSD was adopted. The separation was performed on Diamonsil C18 column (150 mm×4.6mm,5.0μm)with a mixture of acetonitrile - water(32 : 68) as mobile phase and the temperature of column was 35℃. The flow rate was 1.0 mL·min^-1. The temperature of drift tube for measurement was 80℃ ,and the pressure of N2 was 45 psi. RESULTS The standard curve was linear in the range of 7.72 - 23.16μg for astragaloside Ⅳ( r = 0. 9993 ). The average recovery was 97.49% with RSD of 2.08% ( n = 6). CONCLUSION The method is simple, accurate and applicable to control of the quality of Qigui Xiaozheng granule.
