摘  要:本文观察加味四物汤对自发性高血压大鼠(SHR)的心肌组织基因表达谱的影响。提取实验组和对照组SHR心肌组织RNA,用Cy-5、Cy-3逆转录荧光标记制作cDNA探针与表达谱基因芯片杂交,Scan Array4000扫描仪扫描杂交信号荧光强度,ImaGENE3.0软件分析扫描结果,筛选比率在0.5至2.0之间非差异表达基因和比率在0.5到2.0范围之外差异表达基因。结果表明阳性基因重叠有18条,其中高表达的基因有4条,低表达的有14条。该方作用靶点较多,可上调或下调SHR的某些与生物酶、血管活性物质、心肌纤维化、脂肪酸代谢有关基因的表达,这可能是其产生药理作用的重要因素。Tthe effect of supplemented Siwu decoction on cardiac gene expression profile of SHR is observed. Total RNAs were isolated from heart of SHR and those treated with supplemented Siwu decoction. The mRNAs were reversely transcribed to eDNA with incorporation of fluorescent Cy5 - or Cy3 - dUTP to prepare hybridization probes. The mixed probes were hybridized to the cDNA mieroarray. Scan Array 4000 and Ima GENE 3.0 software were used to scan and analyze the fluorescent signals. In the group treated with supple- mented Siwu decoction,4 genes were up -regulation and 14 down -regulation. The decoction has regulatory effect on the gene expression in SHR, which elucidates partly the pharmacologic mechanism in molecular level.
