摘  要:目的探讨p16基因甲基化及蛋白在人非小细胞肺癌组织中的表达情况。方法应用甲基化特异性PCR法(MS-PCR)及免疫组化SP法检测45例非小细胞肺癌组织、21例肺良性疾病组织中p16基因甲基化及蛋白的表达情况。结果肺良性疾病中p16基因甲基化阳性率低于癌组织,差异有显著性(P=0.001),p16基因甲基化与非小细胞肺癌细胞分化程度及淋巴结转移情况有明显的相关性(P<0.05);肺良性疾病中p16蛋白阳性率高于癌组织,差异有显著性(P=0.007);p16蛋白表达与非小细胞肺癌临床分期、细胞分化程度及淋巴结转移情况有明显的相关性(P<0.05)。p16甲基化和p16蛋白表达缺失存在相关性(P=0.004)。结论p16基因甲基化及蛋白的表达情况与非小细胞肺癌的发生、发展有关。Objective To investigate the expression of p16 gene methylation and protein in non - small cell lung cancer. Methods The methylation - specific PCR and SP immunohistochemistry were applied to detect the p16 methylation and protein in tissue of 21 cases of lung benign diseases and 45 cases of non - small cell lung cancer. Results p16 gene methylation rate in non - small cell lung cancer was significantly higher than that in normal lung tissue (P 〈 0. 01 ). The loss rate of p16 gene protein expression in non - small cell lung cancer was significantly higher than that in normal lung tissue (P 〈 0. 01 ). p16 gene methylation was closely correlated with lymph node metastasis and degree of differentiation in non- small cell lung cancer( P 〈 0. 05 ). p16 gene protein expression was correlated with clinical stage, degree of differentiation and lymph node metastasis in non -small cell lung cancer( P 〈 0.05 ). p16 gene methylation was closely correleted with to p16 gene protein expression in non - small cell lung cancer( P 〈 0. 01 ). Conclusion Expression of p16 gene methylation and protein was associated with the carcinogenesis and development of lung cancer.
