摘 要:目的了解脂蛋白脂酶(1ipoprotein lipase,LPL)基因HindⅢ多态性在人群中的分布及其与血脂、体质指数和膳食因素的关系。方法通过聚合酶链反应和限制性片段长度多态性(PCR-RFLP)方法分析比较高脂血症患者156例和健康对照154人LPL-HindⅢ位点多态性,同时进行体格检查、膳食调查及血脂谱项目测定。结果高脂血症组与健康对照组的基因型H+H+、H+H-和H-H-及等位基因H+和H-的构成差异无统计学意义,3种基因型及H+和H-的性别上差异亦无统计学意义;基因型H+H+与非基因型H+H+的血脂谱及BMI水平差异无统计学意义(P>0.05);膳食调查结果显示,高脂血症人群中基因型H+H+与非基因型H+H+的不可溶性膳食纤维摄入量分别为(7.34±3.25)和(8.78±3.22)g,2组比较差异有统计学意义(P<0.05)。结论LPL基因HindⅢ位点非H+H+的不可溶性膳食纤维摄入量较高,此位点多态性不足以构成高脂血症的遗传危险因素。Objective To explore the genotype distribution of lipoprotein lipase polymorphism at Hind Ⅲ locus and its possible association with serum lipid level,body mass index(BMI) and dietary intake.Methods The Hind Ⅲ polymorphism was determined by PCR-PFLP method in 156 adults with hyperlipidemia and 154 adults with normal serum lipid levels in Shanghai.Data of physical examinations,dietary investigation and the levels of serum lipid profile were analyzed for the subjects.Results There was no significant difference in the frequencies of genotypes or allele between the hyperlipidemia and the control,and between male and female.The levels of serum lipid profile and BMI were not significantly different between H+H+ and non-H+H+ genotype groups.The survey of dietary indicated that indissoluble fiber intake differed among the hyperlipidemia with different genotypes.Conclusion The subjects with non-H+H+ had a higher indissoluble fiber intake.While the gene polymorphism at LPL-Hind Ⅲlocus is not indispensable to the genetic risk in hyperlipidemia subjects.
