摘  要:目的探讨丙泊酚对大鼠肢体缺血再灌注后心肌组织损伤有无保护作用。方法制备LIR模型,部分动物在再灌注前经尾静脉给予丙泊酚,光镜下进行心肌组织的形态学观察;采用免疫组织化学法对心肌组织中的肿瘤坏死因子(TNF)-α和核因子(NF)-κB进行定性、半定量检测:用放射免疫法对TNF-α进行定量检测。结果①大鼠肢体缺血再灌注后可致心肌损伤,组织中NF-κB和TNF-α表达升高与对照组比较差异有统计学意义(P<0.05)。②丙泊酚干预后NF-κB和TNF-α表达均减少,与干预前比较差异均有统计学意义(P<0.05)。结论①肢体缺血再灌注组诱发心肌组织损伤;②丙泊酚能降低肢体缺血再灌注组大鼠心肌组织NF-κB和TNF-α的表达,具有抗炎、抗氧化作用,对肢体缺血再灌注诱发心肌组织损害有保护作用。Objective To investigate the effects of Propofol on myocardium injury induced by ischemia-reperfusion. Methods The rat limb ischemia reperfusion model was established, and Propofol was intravascularly injected before reperfusion. The signs of myocardium injury were observed by using microscope, the expression of tumor necrosis factor (TNF)-α and nuclear factor(NF)-κB were qualitatively and semi-quantitatively detected by using immunohistochemical method, method, and the radioimmunoassay was used to detected the TNF-α quantitatively. Results (1) Myocardium injury can be induced by ischemia-reperfusion, and the level of TNF-α and NF-κB were higher in the model group than those in control group (P〈0.05). (2) The level of TNF-α and NF-κB were all declined after implementing the intervention of Propofol, compared with those before intervention (P〈0.05). Conclusion (1) Myocardium injury can be induced by ischemiareperfusion. (2) Propofnl can play a protective role in the myocardium injury induced by ischemia-reperfusion.
