摘  要:目的研究不同饮食模式对大鼠肝脏组织腺苷酸活化蛋白激酶(AMPK)表达及其活性的影响。方法28只雄性SD大鼠适应性喂养1周后随机分为2组:普通对照组(NC组,n=15),高脂饮食组(HF组,n=13)。NC组给予普通饲料,HF组给予高脂饲料。16周后分别测定体重(BW)、血清游离脂肪酸(FFA)、甘油三酯(TG)、总胆固醇(TCH)、空腹血糖(FPG)、空腹胰岛素(FINS)等指标。采用Western blot方法检测两组大鼠肝脏组织中AMPKα和磷酸化腺苷酸活化蛋白激酶α亚基(p-AMPKα)蛋白的水平。结果与NC组相比,HF组大鼠的体重、FFA、TG、FPG、FINS增高(均P<0.05)。两组肝脏组织中AMPKα的蛋白表达水平差异无统计学意义(P>0.05),但HF组肝脏组织中p-AMPKα的蛋白表达水平及AMPK活性降低(均P<0.05)。结论长期高脂饮食减少肝脏组织AMPKα蛋白的磷酸化和AMPK的活性。Objective To investigate the effects of high-fat feeding on the expression and activity of AMPK in liver tissues of rats. Methods 28 male SD rats were randomly divided into two groups and received either normal control diet (NC group,n = 15) or high-fat diet (HF group,n = 13) for 16 weeks. Then all rats'body weight(BW) , free fatty acids ( FFA ), triglyeride ( TG ), total cholesterol ( TCH ), fasting plasma glucose ( FPG ), fasting insulin (FINS) were measured. Protein contents of AMPKα and p-AMPKα in rats liver tissues were determined using Western blot. Results The level of BW, FFA, TG, FPG, FNS were significantly higher in HF rats than that in NC ( P 〈0. 01 ). There were no difference between HF and NC group with regard to the protein expression of AMPKα in liver tissues(P 〉0. 05). But the protein levels of p-AMPKα and activity of AMPK in HF group had a significant decrease ( P 〈 0. 05 ). Conclusion High-fat diet can down-regulate the phosphorylation of AMPK protein contents and activity of AMPK in liver tissues of rats.
