摘  要:目的探讨哺乳期母鼠氯氰菊酯暴露对子代雄鼠成年后精子质量和生育力的影响。方法21只孕鼠被随机分为对照组、低剂量组和高剂量组,于分娩后第1天分别经口灌胃给予母鼠氯氰菊酯[0、6.25、25mg/(kg·d)]处理,染毒至母鼠产后21d,断乳后仔鼠正常喂养至出生后70d,分别评价成年雄性仔鼠精子质量和生育力,TUNEL法检测睾丸细胞凋亡,观察睾丸组织病理学改变。结果与对照组相比,高剂量组附睾精子数明显减少(P<0.05);生育力指标以及睾丸生精细胞凋亡数各组间差异均无统计学意义;组织病理学结果显示,处理组成年雄性后代睾丸生精细胞层减少、管腔内径增大、细胞相对稀疏且排列较紊乱。结论哺乳期母鼠氯氰菊酯暴露对雄性子代成年后精子质量有明显影响。Objective To explore the effects of maternal cypermethrin exposure during lactation on sperm quality in adult male offspring in mice. Methods Twenty-one pregnant mice were randomly divided into three groups. Maternal mice in cypermethrin groups were orally administered with different doses of cypermethrin [ 0,6.25,25 mg/( kg·d) ] daily from postnatal day 1 (PND1) to PND21. Sperm quality and fertility were evaluated in adult male offspring at PND70. Testicular apoptosis was detected by TdT-mediated dUTP nick end labeling (TUNEL). Histopathological changes in testicular tissues were observed. Results There was a significant decrease in the number of epididymal spermatozoa in male offspring of high-dose group as compared with controls ( P 〈 0. 05 ). Histological examination showed that maternal cypermethrin exposure during lactation significantly increased the inside diameter (ID) of seminiferous tubules, decreased the number and layers of spermatogenic ceils, and disturbed the array of spermatogenic cells in male pups at PND70. However, there was no difference in fertility of adult male offspring among different groups. Conclusion Maternal cypermethrin exposure during lactation can influence sperm quality of adult male offspring.
