摘  要:目的研究下肢缺血预处理对急性心肌梗死大鼠心肌的保护作用。方法随机将大鼠分成急性心肌梗死组(AMI)、下肢缺血预处理组(LIP)和正常对照组(NC)3组,每组15只。各组大鼠分别在急性心肌梗死后处死取材。经冷冻、切片后用2%TTC法染色,于手术显微镜下仔细分离梗死部位(IS)和缺血危险区(AAR),并计算IS与AAR重量比值(IS/AAR),推测心肌梗死面积。测定各组大鼠心肌组织中血红素氧合酶-1(HO-1)活性、NO水平、心肌组织丙二醛(MDA)、超氧化物歧化酶(SOD)含量。结果LIP组HO-1活性升高,心肌梗死面积比心梗组明显减小;LIP组心肌组织MDA和NO水平显著低于AMI组,差异有统计学意义(P<0.05)。结论下肢缺血预处理可使心肌内源性保护因子HO-1升高,可通过减少NO生成和抗氧化作用,对心肌产生保护作用。Objective Studied the protective role of the lower limbs ischemic preconditioning(LIP) on acute myocardial infarction of rat.Methods Sprague-dawley(SD) rats were divided into acute myocardial infarction group,the lower limbs ischemic preconditioning group and normal control group.All the rats were sacrificed after AMI.Used trephine tetrazolium chloride(TTC) staining after deepfrozed and sliced,was carefully isolated under a microscope,surgical infarct area(IS) and ischemic risk area(AAR),and calculated ratio of IS and AAR weights (IS/AAR) , specuiated that myocardial infarct size. Determined the heine oxygenase(HO-1)activity,NO levels,myocardial malondialdehyde(MDA) ,superoxide dismutase(SOD) levels of myocardial tissue of rats in each group. Results The enzymatic activity of HO-1 was step up in LIP,the myocardial infarct area was decreased in LIP that compared with group AMI, myocardial tissue MDA and NO levels in LIP group were significantly lower than that in AMI group, the differences were statistically significant ( P 〈 0.05 ). Conclusion Lower limb ischemic preconditioning could protected the myocardium-derived factor HO-1 increased by reducing NO production and anti-oxidation, to produce a protective effect on the myocardium.
