摘 要:目的研究甘草次酸(GA)对中药活性成分与牛血清白蛋白(BSA)结合的影响。方法利用多种光谱技术和毛细管电泳前沿分析方法测定,用Stern-Volmer方程和Scatchard plot等对数据进行处理。结果得到甘草次酸与BSA的结合常数、位点数等结合特征,中药活性成分与BSA结合的常数和甘草次酸对这些成分结合常数的影响。结论甘草次酸与牛血清蛋白有高、低两类结合位点,总的结合位点数为8,蛋白与GA浓度之比为0.38时,结合率已高达95%。GA使中药活性成分与牛血清蛋白的结合常数下降最大达67%,可能对它们的药理作用产生影响。Objective To evaluate the influence of glycylwhetinic acid (GA) on the binding of active ingredients of Chinese herbs with serum albumin. Methods Spectroscopic method and eapillaly eleetrophoresis frontal analysis were utilized to determine the binding data. Stern - Volmer equation and Scatchard plot were used to process the data. Results Binding constant and sites etc of GA with BSA were got. Binding eonstants of active ingredients of Chinese herbs and influence of GA on the binding of them were aslo obtained. Conclusion There are two kinds of binding sites (high and low) in BSA while binding with GA and the total binding sites are eight. The binding percentages of GA are up to 95% when the ratio of BSA to GA is 0.38. In the presence of GA, binding constants of active ingredients of Chinese herbs decrease up to 67% , which may exert an influence on their pharmacological action
