加味失笑散对家兔心肌缺血再灌注损伤保护作用机制的研究

摘　　要：目的研究加味失笑散对心肌缺血再灌注损伤保护的作用机理。方法将日本大耳白家兔40只随机分为5组:假手术组、模型组、加味失笑散低剂量组、加味失笑散中剂量组和加味失笑散高剂量组。采用结扎左冠状动脉前降支的方法制备缺血和再灌注模型,通过检测缺血前、缺血30min和再灌注60min时血清中超氧化物歧化酶(SOD)、丙二醛(MDA)和血清肌酸激酶(CK)的变化,探讨加味失笑散对心肌缺血再灌注损伤保护作用的机制。结果缺血30min和再灌注60min时模型组实验动物的SOD活力较假手术组明显降低而MDA和CK明显增高(P<0.01),各用药组动物血清中的SOD活力明显高于模型组,MDA和CK活力明显低于模型组(P<0.01),且呈明显的剂量依赖性。结论加味失笑散对心肌缺血再灌注的损伤有保护作用,其作用机制是通过增强SOD的活力,清除自由基而减轻心肌的损伤。Objective To study the protective mechanism of Jiawel Shixiaosan in isocheimmia - reperfusion injury, of myocardia in rabbits. Methods Forty rabbits were divided randomly into 5grunps： sham operation group, model group, low dose group of Jiawei Shixiaosan, middle dose group of Jiawei Shixiaosan and high dose group. The isocheimmia and isocheimmiareperfusion models were made by ligating the left anterior descending branch of coronary artery. SOD, MDA and Ck in serum of the experimental rabbits were detected separately before isocheimmia, after isoeheimmia 30min and reperfusion 60min. Results In 30min after isoe t heimmia and 60min after reperfusion the activities of the serum SOD in the model group were decreased significantly compared with those in sham operation, and the serum MDA, CK in the model group were increased significantly compared with those in sham operation. The activities of the serum SOD in every experimental group given Jiawei Shixiaosan were significantly higher than those in model group, and the serum MDA, CK in the experimental group given Jiawei Shixiaosan were significantly lower than those in model group. The change of the content of the serum SOD, MDA, CK were significantly associated with the close of Jiawei Shixiaosan. Conclusion Jiawei Shixiaosan has protective effect against ischemia - reperfusion injury of myoeardia in rabbits through reducing the activity of SOD and removing oxyradical.
