摘  要:目的研究加水量、煎煮时间、煎煮次数、浓缩条件等水煎工艺控制因素对附子毒效组分含量相关性的影响。方法以生附子和常用炮制品为对照,采用离子对分光光度度法与异羟肟酸铁法分别测定总生物碱(TA)、酯型生物碱(EA)组分含量,并以毒效因子[W=(TA-EA)/TA]为综合评价指标,研究工艺因素对毒效因子的影响。结果不同炮制品和生附子药材毒效因子差别较大;水煎过程中,煎煮条件对生品影响较大,黑顺片的毒效因子基本保持稳定为97%左右,白顺片为81%,煎煮时间和次数是主要影响因素。结论炮制品水煎过程毒效因子的相对稳定可能是附子汤剂发挥药效、安全使用的先决条件,毒效组分不同比例可能是附子不同炮制品发挥不同药效的物质基础。Objective To research the effect on the Aconite toxicodynamics component's content related to decoction process controlling factors such as water volume, boiling time, boiling numbers, concentrated decoction process. Methods With CRALP (Crude Radix aconiti lateralis preparata)and common processed products as a control, ion-pair spectrophotometry and acid dye colorimetry and hydroxamic acid iron spectrophotometly were used to determine the content of total alkaloids and ester alkaloids, and toxicodynamics factor [ W = (TA-EA) / TA] for the comprehensive evaluation index to study the process factors on the impact of toxicodynamics factor. Results Different processed products and CRALP showed obvious differences in toxicodynamics factor. In water decoction process, decocting conditions showed great impact on CRALP,the toxicodynamics factor of BPLRA( Black prepared lateral root of aconite) basically remained stable for about 97%, and WPLRA (White prepared lateral root of aconite) 81%. Decoeted time and times were the main factors. Conclusion Decoction toxicodynamics factor relative stability may exert a prerequisite for the effect and safe use ; different proportions of toxicodynamics components may be material basis of different pro- cessed products.
