摘　　要：目的研究3-甲氧基-4-O-(2’-亚甲基-3’5’6’-三甲基吡嗪基)阿魏酸钠(MC I-002)对大鼠局灶性脑缺血的保护作用及机制。方法应用大鼠局灶性脑缺血损伤模型,观察MC I-002对脑缺血大鼠神经行为、脑含水量、脑梗死率、病理组织学以及组织内超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)、丙二醛(MDA)、血栓烷B2(TXB2)和6-酮-前列腺素(6-Keto-PG I2)的影响。结果MCI-002可改善局灶性脑缺血大鼠的神经行为,降低脑含水量和脑梗死率,改善其病理组织学,显著升高6-keto-PGF1α/TXB2比值、提高SOD活力、降低MDA含量且呈剂量依赖性。结论MCI-002对大鼠局灶性脑缺血有很好的保护作用,其机制可能与调节PG I2/TXA2比值,改善能量代谢和抗氧化有关。Aim To study the protective effect and its mechanism of MCI-002,3-methoxy-4-O- （ 22methyl-35 6-Trimethylpyrazine） ferulic sodium,on focal cerebral ischemia in rats. Methods Neurological behavior,the pathologic change,the cerebral water volume,the cere- bral infarction rate,the cerebral biochemistry of cerebral ischemia rats such as activity of SOD,LDH,TXB2 ,6-Keto-PGI2 and content of MDA were evaluated using the rat model of focal cerebral ischemia. Results MCI-002 improved the neurological behavior, decreased the cerebral water volume and cerebral infarction rate,enhanced the LDH and SOD activity,lowered the MDA content,increased the 6-keto-PGF1 α/TXB2 ratio, and detained the pathologic change of ischemic brain in a dose-dependent way. Conclusion MCI-002 has significantly protective effect against focal cerebral ischemia in rats, and its mechanism may be related to its improvement of energy metabolism and anti-oxidation.
