摘  要:以狗牙根的嫩叶为材料,在传统CTAB法的基础上加以改良,建立了狗牙根基因组DNA的CTAB提取方法。得到的DNA经核酸蛋白质分析仪测得A260/A280值处于1.69~2.02之间、A260/A230集中处于1.82~2.08之间,经琼脂糖凝胶电泳检测各样品具有一条明亮主带并无拖尾现象,经ISSR-PCR检测出现清晰稳定的条带,说明改良CTAB法提取得到的DNA完整性好,纯度高,适用于分子标记等分子生物学的研究。The extraction of genomic DNA from Cynodon dactylon is a precondition of molecular biological research. An improved method of extracting genomic DNA from young leaves of Cynodon dactylon was established. The obtained DNA was detected by nucleic acid and protein analyzer and the results showed that A26o/A~ was from 1.69 to 2.02 and A^o/A^0 concentrated from 1.82 to 2.08; the extracted products had a bright band without tailing which could be observed through agarose gel electrophoresis and it also had stable and bright bands which were detected by ISSR-PCR. These results indicated that the genomic DNA extracted by improved CTAB method had good integrality and high purity, and it could be used in each kinds of molecular biological research.
