胃癌组织中心体相关激酶Nek2的表达及其意义

摘　　要：目的:研究中心体相关激酶Nek2(Nek2)在胃癌组织中的表达,为胃癌的诊断和治疗提供新的靶点.方法:收集外科手术切除的胃癌组织标本及胃切缘组织标本50例,采用实时荧光定量逆转录聚合酶链反应(RT-PCR)、免疫组织化学SABC法检测胃癌组织及远癌切端胃组织Nek2的表达情况,分析Nek2表达与临床病理参数之间的关系.结果:胃癌组织Nek2mRNA表达水平(2.29±1.67)显著高于远癌切端胃组织(1.26±0.87),差异具有统计学意义(P<0.05).胃癌组织Nek2mRNA和蛋白表达水平在不同浸润深度、TNM分期、肿瘤分化程度、有无淋巴结转移组中的差异均具有显著性,其中T3+T4组、Ⅲ+Ⅳ期组、分化较低组和有淋巴结转移组分别显著高于T1+T2组、Ⅰ+Ⅱ期组、分化较好组和无淋巴结转移组,差异具有统计学意义(P<0.05).结论:胃癌组织中Nek2分子表达上调,且与肿瘤浸润深度、分化程度、TNM分期、淋巴结转移相关,提示其参与胃癌的发生发展过程,可作为新的胃癌诊断及判断预后的指标.AIM：To analyze its effect on the invasionof gastric cancer and to find a new target for clinical diagnosis and treatment of gastric cancer by investigating the expression of centrosome-relatedkinase-Nek2 in gastric cancer.METHODS：Gastric cancer tissues and tissues in the surgical margin were collected from 50 patients who underwent surgery in our hospital.Real time quantitative reverse transcriptase polymerase chain reaction and immunohistochemistry were used to detect the expression of Nek2 in these samples.The correlation between the expression of Nek2 and patients＇ clinicopathologic parameters were analyzed.RESULTS：The expression of Nek2 mRNA and protein in gastric cancer were significantly higher than that in tissues in the surgical margin（P0.05）.There was significant correlation between the expression of Nek2 mRNA and protein in gastric cancer and TNM stage,depth of primary tumor invasion,differentiation and lymph node metastasis（P0.05）.CONCLUSION：The Nek2 is overexpressed in gastric cancer at mRNA and protein levels,and positively correlated with the depth of primary tumor invasion,differentiation,lymphatic node metastasis as well as TNM stage of gastric cancer,which indicates that Nek2 promotes tumor progression to some degree.Nek2 can be used to determine the extent of progress and prognosis of gastric cancer.
