菊藤胶囊对自发性高血压大鼠炎症因子的影响

摘　　要：目的:观察心脏组织及血清中肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)的表达,探讨中药菊藤胶囊抗高血压的炎症作用机制.方法:采用自发性高血压大鼠(SHR)模型,用大鼠血压测定仪测定大鼠尾动脉压;取各组大鼠血采用ELISA法检测血清TNF-α,CRP水平;免疫组织化学法检测心脏组织TNF-α,CRP的表达.结果:SHR给予药物干预前,各组血压相当,4wk后空白对照组血压明显升高;与空白对照组相比较,菊藤胶囊组和卡托普利组均能降低血压(P<0.05),而菊藤胶囊组与卡托普利组的血压水平相当.血清ELISA测定结果显示,CRP菊藤胶囊组(27.8±12.0)ng/L较卡托普利组(48.7±12.9)ng/L水平降低(P<0.05),2组与空白对照组(71.3±16.4)ng/L相比较均显著降低(P<0.05);TNF-α菊藤胶囊组(22.5±8.3)ng/L较卡托普利组(24.0±9.2)ng/L差异无显著性,2组与空白对照组(39.3±16.7)ng/L相比较均显著降低(P<0.05).心脏组织免疫组化观察显示其阳性表达特异性不强与实验周期过短有关,但参照对比观察显示,心肌细胞阳性棕黄色颗粒空白对照组密布于组织上,染色较深;卡托普利组较少且染色较浅;菊藤胶囊组部分散在阳性着色,染色较淡.菊藤胶囊组、卡托普利组与空白对照组相比较TNF-α,CRP的表达均降低.结论:菊藤胶囊可以降低血压和SHR血清TNF-α,CRP水平,下调心肌组织的TNF-α,CRP表达,减少炎症反应引起的心肌损害.AIM：To study the effect and machanism of Juteng Capsule in treating spontaneous hypertensive rats through mesuring blood pressure and observing the expression of TNF-α and CRP so as to provide theoretic basis of inflammatory function for the clinical application of Juteng Capsule.METHODS：A total of 30 male SHR rats were randomly divided into 3 groups（n=10）：blank group,Captopril and Juteng group.The tail artery blood pressure was determined by using instrument for measuring blood pressure in rats.the whole blood was collected and the serum level of TNF-α and CRP was measured by ELISA in each group,and the expression of TNF-α and CRP in myocardium was observed with the immunohistochemicalstaining in each group.RESULTS：After 4 weeks of drug therapy,the blood pressure was significantly decreasedin Juteng and Captopril group compared with that in blank group（P0.05）;the contents of TNF-α and CRP in serum were decreased in Juteng and Captopril group compared with that in blank group（P0.05）,significant difference appeared in Juteng group compared with that in Captopril group（P0.05）;immunohistochemistry examination of the myocardium showed that the expression of TNF-α and CRP in Juteng and Captopril group was lower than those of in the blank group.CONCLUSION：Juteng Capsule can lower blood pressure,degrade the level of TNF-α and CRP in serum,reduce the expression of TNF-α and CRP in myocardium,decrease cardiac damage due to inflammatory reaction.
