摘 要:用等电聚焦电泳(IEF)和Western-blot技术检测125例Ⅱ型糖尿病患者血清apoA-Ⅳ的基因频率、表型分布和血清水平,以了解Ⅱ型糖尿病患者apoA-Ⅳ的多态性及其与apoA-Ⅳ血清水平的关系。结果显示:Ⅱ型糖尿病患者apoA-Ⅳ的两个常见等位基因频率为:apoA-Ⅳ*1=0.9960,apoA-Ⅳ*2=0;两个常见表型的分布为:apoA-Ⅳ1-1=99.2%,apoA-Ⅳ2-1=0;apoA-Ⅳ的血清水平为0.0262±0.0125g/L,与健康人群组相比较,apoA-Ⅳ多态性没有显著性差异(P>0.75,P>0.25),其血清水平显著增高(P<0.001)。结论;Ⅱ型糖尿病患者apoA-Ⅳ的多态性不是发病的决定因素。To study apoA-Ⅳ polymorphism and its relation to serum isoelectric focusing (IEF) and immuno-apoA-Ⅳ level in type 2 diabetes patients,serum apoA-Ⅳ level,gene frequency and phenotype distribution were determined by blotting technique in patients from Hangzhou. The results showed that the frequency of apoA-Ⅳ. 1 and apoA-Ⅳ. 2,two common alleles was 0. 996 and 0 respectively. The distribution of apoA-Ⅳ 1-1 and apoA-Ⅳ 2-1, the two common phenotypes was 99. 2 % and 0 respectively. The serum level of the apoA-Ⅳ in type 2 diabetes was 26. 2 ±12. 5mg/dl. No statistically significant difference was seen in apoA-Ⅳ allele frequencies and phenotype distribution between normal Chinese and type 2 diabetic patients. Serums apoA-Ⅳ concentration was significantly higher in diabetes than in normal individuals(P <0.001). It is concluded that the apoA-Ⅳ polymorphism is similar in the two populations and that the apoA-Ⅳ polymorphism has no significant impact on serum apoA-Ⅳ level.
