摘　　要：目的制备抗EOLA1多克隆抗体并检测其在肿瘤组织中的表达。方法采用原核表达获得EOLA1蛋白,纯化后作为抗原免疫大白兔,获得兔抗人EOLA1(hEOLA1)多抗血清,进行免疫组化检测人多肿瘤芯片上EOLA1的表达。结果制备了高滴度的hEOLA1多克隆抗体,免疫组化显示EOLA1蛋白在肺癌、结肠癌、胃癌和肝癌仅表达于肿瘤间质,肿瘤细胞本身未见表达,而在胶质瘤和乳腺癌则呈广泛表达。结论EOLA1可能在胶质瘤和乳腺癌的发生、发展方面起着重要作用。Objective To prepare the anti-endothelial-overexpressed lipopolysaccharide-associated factor 1 （EOLA1） antibody and detect its expression in tumor tissues. Methods Purified EOLA1 protein produced by prokaryotic expression,was used to immunize white rabbit, and the anti-EOLA1 polyclonal antibodies were collected from the rabbit serum and identified by Western blotting. Then the antibodies were used for immunohistochemical assay in human poly tumor chip. Results The antibody we generated was of very high titer, and could be used for detection of EOLA1 protein by Western blotting. In immunohistochemical assay, EOLAI protein was expressed only in the interstitial tissues of lung, colon, stomach and liver tumors. However, in glioma and mammary cancer EOLAI protein was expressed in both the interstitial and tumor cells. Conclusion EOLA1 may be play a important role in the genesis and development of glioma and mammary cancer.
