摘 要:目的探讨补肾活血中药对雷公藤致卵巢储备下降大鼠颗粒细胞凋亡的影响。方法 12周龄雌性SD大鼠60只,随机分为6组:空白组、雷公藤模型组、补肾活血方低、中、高剂量组及对照药六味地黄方组。观察灌服上述不同药物后大鼠一般情况、卵巢指数、卵巢组织病理形态、血清E_2、FSH水平及卵巢颗粒细胞凋亡因子Bcl-2、Bax、Caspase-3蛋白的表达。结果 Bcl-2、Caspase-3表达:模型组、补肾活血方低剂量组与空白组比较有极显著性差异或显著性差异(p<0.01或P<0.05);六味地黄方组和补肾活血方低、中、高剂量组与模型组比较均有极显著性差异或显著性差异(P<0.01或P<0.05)。Bax表达:模型组、补肾活血方低剂量组与空白组比较有极显著性差异(P<0.01);六味地黄方组和补肾活血方中、高剂量组与模型组比较有极显著性差异或显著性差异(P<0.01或P<0.05)。结论细胞凋亡调节因子Bcl-2、Bax、Caspase-3蛋白与卵巢储备密切相关,补肾活血中药抑制卵巢颗粒细胞凋亡、减少卵泡闭锁是其改善卵巢储备功能的可能机制。Objective To discuss the effects of BuShenHuoXue on ovarian granulose cell apoptosis of DOS rats induced by GTW. Methods Sixty 12 - week - old female SD rats were randomly divided into 6 groups: control, model GTW,low dose of BuShenHuoXue Formula, middle dose of BuShenHuoXue Formula,high dose of BuShenHuoXue Formula, and LiuWeiDiHuang soup group. The ovary index, pathomorphological configuration, the level of E2 and FSH in blood serum, and the expression of granular cell apoptosis protein Bcl - 2, Bax and Caspase - 3 was obserred after giving rats the different drugs separately. Results The expression of Bcl - 2, Caspase - 3 : as compared with control, the model, BuShenHuoXue low dose had a significant difference (P〈0.01 or P〈0.05) ;as compared with the model LiuWeiDiHuang soup,the low dose of BuShenHuoXue,the middle dose of BuShenHuoXue and the high dose of BuShenHuoXue had a very significant difference ( P 〈 0.01 or P 〈 O. 05). The expression of Bax: as compared with control, the model and the low dose of BuShen- HuoXue had a very significant difference( P 〈 0.0l );as compared with the model , LiuWeiDiHuang soup, the middle dose of BuShen- HuoXue and the high dose of BuShenHuoXue had a very significant difference (P 〈 0.01 or P 〈 0.05). Conclusion Apoptosis regulator Bcl -2,Bax and Caspase -3 protein are closely related with the ovarian reserve. The inhibition of ovary apoptosis and the reduction of follicular atresia are potential mechanism of improvement of ovary reservation.
