薄层扫描法测定血浆中盐酸氟桂利嗪浓度及药物动力学

摘　　要：血浆中盐酸氟桂利嗪经硼酸缓冲液酸化后，正戊烷—异丙醇（98：2）提取其原型，加酸使成盐溶于无机相中，再加碱后用二氯甲烷提取，样品点于硅胶GF_(254)薄层板上，以环己烷—丙酮—氯仿（9：3：5）为展开剂．氟桂利嗪在254nm紫外灯下呈紫色斑点，Rf＝0．54．于CS－930薄层扫描仪上测定，λs＝215nm、λR＝310nm，线性范围0．3～8μg，平均回收率90．02％．用此法测定了氟桂利嗪血药浓度，并进行药物动力学研究．Plasma samples were acidified by Broate buffer solution and extracted by n - pentane- 2 - propanol (98: 2). Added acid, the drug was redissolved into inorganic phase and then alkalized and extracted by dichloromethane. The extracted solution was spotted on silica gel F254 plate and developed with cyclohexane-ketone-chloroform (9: 3: 5). Under UV 254 nm lamp, flunarizine shows violet spot with Rf value about 0. 54.scanning at CS- 930 TCL scanner λS=215 nm; λm=310 nm. Liner range was between 0. 3~8 ug, with average recovery 90. 02%. The concentration of the drug in plasma and the Pharmacokinetics were studied by the proce-dure.
