摘  要:目的本研究分析延边汉族和朝鲜族人群中脂联素基因启动子的单核苷酸多态性(SNPs)与2型糖尿病的关联。方法共有97个延边个体被纳入到本研究中,包括55个汉族(13个正常人和42个2型糖尿病患者)和42个朝鲜族(16个正常人和26个2型糖尿病患者)。检测项目包括体重指数(BMI)、空腹血糖(FPG)、总胆固醇(TC)和甘油三酯(TG)。PCR和测序用于SNP的筛选。ANOVA和X2检验用于数据分析。结果在延边朝鲜族中TG在2型糖尿病组大于正常组(P=0.033)。FPG、TC和TG在延边朝鲜族2型糖尿病组大于延边汉族2型糖尿病组(P=0.038;P=0.037;P=0.047)。在延边朝鲜族中-11426A-11377C的单体型频率在2型糖尿病组小于正常组(P=0.043)。在延边汉族中BM]与-11426位点的基因型有关,基因型-11426G的BMI小于基因型-11426A(P=0.039)。-11426位点AA基因型的BMI在延边汉族大于延边朝鲜族(P=0.006)。结论1.延边汉族和朝鲜族之间在2型糖尿病组中FPG、TC和TG存在民族差异。2.在延边朝鲜族中单体型-11426A-11377C可能在2型糖尿病发展过程中被认为是-种保护因素。3-在延边汉族中SNP-11426A〉G可能与肥胖有关。Objective This study was designed to analyze the possible association between single nucleotide polymorphisms (SNPs) within the promoter of adiponectin gene and type 2 diabetes in Yanbian Han and Korean Chinese populations. Methods We enrolled a total of 97 subjects from Yanbian, including 55 Han Chinese ( 13 healthy individuals and 42 type 2 diabetes patients) and 42 Korean Chinese ( 16 healthy individuals and 26 type 2 diabetes patients). Body mass index (BMI), fasting plasma glucose (FPG), total cholesterol (TC), and triglyceride (TG) were measured. PCR and sequencing were used for screening of SNPs of promoter of adiponectin gene. ANOVA and X2 test were used for analysis of data. Results The level of TG in type 2 diabetes group was higher than that in normal group in Yanbian Korean Chinese (P = 0. 033). The level of FPG, TC, and TG in type 2 diabetes group of Yanbian Korean Chinese were higher than those in type 2 diabetes in Yanbian Han Chinese (P = 0. 038, P = 0. 037, and P = 0. 047, respectively). The frequency of haplotype of - 11426A - 11377C in type 2 diabetes group was less than in normal group in Yanbian Korean Chinese (P = 0. 043 ). In Yanbian Han Chinese, BMI was relative to the genotype at locus of - 11426, and the level was lower in individuals with allele - 11426G than that with -11426A (P = 0. 039). And the BMI level with genotype of AA at locus of -11426 in Yanbian Hart Chinese was higher than it in Yanbian Korean Chinese (P = 0. 006). Conclusion Three conclusions can be drawn. First, Yanbian Han and Korean Chinese had national difference in FPG, TC, and TG in type 2 diabetes group. Second, the haplotype of - 11426A - 11377C could be considered as a protective factor in the progression of type 2 diabetes in Yanbian Korean Chinese. Third, the SNP of -11426A 〉 G might be associated with obesity in Yanbian Han Chinese.
