摘 要:目的系统研究绞股蓝抗诱变作用及剂量效应。方法采用鼠伤寒沙门氏菌营养缺陷型回复突变试验(A-m es试验)、人外周血淋巴细胞姐妹染色单体交换(SCE)试验、人外周血淋巴细胞微核(MN)试验、中国仓鼠卵巢细胞(CHO)染色体畸变试验,对绞股蓝水提物低、中、高剂量组的抗诱变性能进行系统研究。结果绞股蓝低、中、高剂量组对2,7-AF诱发的TA98回变菌落数增加效应均表现出抑制作用,高剂量组作用十分显著(P<0.01),抑制率达到58.0%;对NaN3诱发的TA100回变菌落增加效应,绞股蓝中、高剂量组也具有抗诱变作用,高剂量组作用显著(P<0.01),抑制率为47.2%。绞股蓝中、高剂量组能抑制由MMC诱发,淋巴细胞SCE率的增高(P<0.05);绞股蓝三个剂量组均能明显降低由MMC诱发人淋巴细胞MN率的增高,中、高剂量组效果十分明显(P<0.01)。绞股蓝三个剂量组均能降低由MMC诱发的CHO染色体畸变率,各组间表现出明显的剂量-效应关系(P<0.05),且中、高剂量组作用显著(P<0.01),高剂量组抑制率达到49.15%。结论绞股蓝在遗传毒理学的体外测试系统中表现出较强的抗诱变性。objective To accurately determine the anti - mutation of the aqueous extract of Gynostemma pentaphyllum. Methods They used world - wide adopted means [ Amos test, Sister chromatid exchangos test in the lymphocytes of human blood, Micronucleus test in the lymphocytos of human blood, Chromosomal aberration test of CHO cells] to systematically study the anti - mutation of Gynostemma under different doses. Results: All three dose groups for Gynostemma pentaphyllum can restrain the increase of the back matagenesis of TA98 induced by 2,7 -AF, high dose groups are especially effective (P〈0.01) , a inhibition rate of 58.0% respectively. Median dose group can restrain the increase of the back matagenesis of TA100 induced by NaN3 ( P 〈 0. 05 ), high dose group shows significant effect (P 〈 0.01,47.2% inhibition rate). High and Median dose group inhibits the increase of SCE] in the lymphecytos of human blood induced by MMC (P 〈0.05). All three dose groups for Gynostemma pentaphyUum can restrain the increase of MN in the lymphocytos of human blood induced by MMC, high and median dose groups are especially effective (P 〈0.01 ). High does group for Ginseng inhibits the Chromosomal aberration of CHO cells induced by MMC. While all three dose groups for Gynostemma pentaphyllum inhibit the Chromosomal aberration of CHO cells induced by MMC, and show clear dose-effect relationship (P〈0.05). The high dose groups is especially effective (P〈0.01), it show inhibition rate 49.15% respectively. Conclusions Gynostemma pentaphyllum show good anti - mutant activity in vitro genetic toxicology test systems.
