多沙唑嗪对犬的长期毒性实验研究

摘　　要：目的：在动物体内验证多沙唑嗪临床毒副反应及性质。方法：以多沙唑嗪１，４，１６ｍｇ／ｋｇ，给犬连续９０ｄ，ｐｏ，并设对照组，观察用药期间毒性反应及停药２１ｄ期间恢复情况。结果：高剂量组在给药初期动物出现眼睑下垂、结膜及巩膜充血、瞬膜弛缓、眼分泌增多等现象，部分动物活动量减少，当继续用药１０～２０ｄ后症状逐渐减轻并消失。在用药６３ｄ后，动物体重增长明显减慢（Ｐ＜０．０５），且在６０ｄ及９０ｄＢＵＮ含量明显增高（Ｐ＜０．０５）。停药后均恢复正常。中、低剂量组未见异常现象。结论：多沙唑嗪对犬的无毒反应剂量大约为４ｍｇ／ｋｇ。OBJECTIVE:To verify the clinical toxicity and adverse effect of doxazosin by animals.METHODS:Dogs were treated with oral doxazosin in the regimens of 1,4 and 16 mg/kg for 90 days,and the toxic reactions during treatment and the recovery condition within 21 day of stopping treatment were observed.RESULTS:In the high dose group,blepharoptosis,conjunctival and scleral congestion,relaxation in nictating membrane,and increase in tear secretion in all dogs and slow in activity in some dogs were observed in the early stage of treatment,but these symptoms gradually disappeared in 10～20 days of continuous dosing.The body weight of dogs in the high dose group reduced significantly after 63 days of treatment.The laboratory test indicated that the BUN content increased significantly in dogs of the high dose group.No obvious changes in other groups were observed.All the toxic symptoms restored and disappeared after withdrawal of treatment.CONCLUSION:The safe dosage of doxazosin for dogs is about 4 mg/kg.
