摘  要:目的观察卵清蛋白(OVA)诱导建立的急性支气管哮喘大鼠模型,采用外源性硫氢化钠(NaHS)处理后Th1/Th2细胞因子及肺部黏蛋白基因Muc5ac的表达情况,探讨气体信号分子硫化氢(H2S)在哮喘发病中的作用。方法26只健康SD大鼠按照随机数字表法分为正常对照组(A组)、哮喘组(B组)和NaHS干预组(C组),每组分别为8只、9只、9只。激发24h后解剖大鼠,取腹腔静脉血检测其IL-4及IFN水平。取肺组织观察大鼠支气管周围的炎性细胞浸润程度并进行评分;采用免疫组织化学染色法测定大鼠肺中Muc5ac的表达情况。结果光镜下哮喘大鼠支气管周围炎性细胞浸润程度评分(采用中位数表示)A组1分,B组4分,C组2分,3组间比较差异有统计学意义(H=13.75P<0.05)。A、B、C组血浆IL-4水平分别为(99.25±31.60)、(185.40±139.30)和(262.00±27.10)pg/L,3组间比较差异有统计学意义(F=7.706P<0.05)。A、B、C组IFN-γ水平分别为(177.2±73.0)、(91.8±32.3)和(120.7±31.5)pg/L,3组间比较差异有统计学意义(F=6.731P<0.05)。A、B、C组肺Muc5ac蛋白水平(用阳性面积表示)分别为(501.75±270.58)、(1330.22±662.87)和(715.88±282.49)μm2,3组间比较差异有统计学意义(F=11.82P<0.01)。结论哮喘大鼠黏蛋白基因Muc5ac表达增加,外源性H2S可减轻哮喘呼吸道炎性反应,对哮喘急性发作起保护作用。Objective To explore the effect of gaseous signal molecule hydrogen sulfide ( H2S) on the expression of lung Muc5ac and Th1/Th2 eytokines in ovalbumin(OVA) - induced asthma rats and explore the effect of H2S on asthma. Methods Twenty - six healthy SD rats were randomly divided into 3 groups : control group ( Group A, n = 8), asthma group ( Group B,OVA induced, n = 9) and NariS group ( Group C, OVA induced rats treated with NariS, n = 9 ). Twenty - four hours after treatment, rats were anatomized to measure serum interleukin (IL) -4, interferon - γ and the levels of infiltration into inflammatory cells around bronchus were observed, which were scored with the optical microscope. The expression of lung Muc5ac was determined by immunohistochemical staining. Results The score of the levels of infiltration into inflammatory cells around bronchus expressed by median was 1 seore in group A, 4 score in group B and 2 score in group C. There were dramatically statistics significance among the 3 groups (H = 13.75 P 〈 0.05 ). The level of serum IL - 4 was (99.25 ± 31.60) , ( 185. 40 ± 139.30) and (262.00 ± 27. 10) pg/L, respectively in group A, B and C, there were dramatically statistical significance among the 3 groups(F=7.706 P〈0.05).The level of serum IFN -γ was (177.2±73.0),(91.8±32.3) and (120.7±31.5) pg/L,reseparately in group A, B and C, there were dramatically statistical significance among the 3 groups ( F = 6. 731 P 〈 0.05 ). The expression of lung Mue5ac, expressed with positive areas, were (501.75 ±270.58) ,(1 330.22 ±662.87) and (715.88 ±282.49) μm^2 ,respectively in group A, B and C,there were dramatically statistical significance among the 3 groups (F = 11.82 P 〈 0.01 ). Conclusions The expressions of lung Muc5ac increase in OVA -induced asthma rats. Exogenous H2S may play an important role in alleviate airway inflammation and protection from asthma acute break out.
