摘 要:[目的]探讨苦杏仁苷促进人血T淋巴细胞早熟凝集染色体(PCC)分裂增殖的作用,为辐射损伤快速生物剂量估算提供依据。[方法]每一实验项目取6名不同实验者的周围血加入不同浓度的苦杏仁苷进行实验。姐妹染色单体互换(SCE)方法用于观察苦杏仁苷最佳增殖浓度和促进细胞进入下一分裂周期的作用,以第2代细胞率为指标;常规的PCC方法(培养48h)和本研究的PCC方法(培养40h、加不同浓度苦杏仁苷)用于观察PCC细胞的G1、S、G2和M期,以G2+M期PCC细胞率作评价指标。[结果]①每一实验者苦杏仁苷均明显促进T淋巴细胞的增殖(P<0.01~P<0.005),最佳浓度为每L培养体系含200mg苦杏仁苷,比0mg/L的第2代细胞率增加0.74~1.60倍。②以苦杏仁苷促进细胞增殖的最佳浓度培养体系,培养28h加入秋水仙素的时间点出现第2代细胞(5‰~9‰);而未加苦杏仁苷的样本要在培养36h加秋水仙才出现第2代细胞(4‰~9‰)提早8h。③以苦杏仁苷最佳浓度配合花萼海绵体诱癌素A的培养体系培养,获得T淋巴细胞G2+M期的PCC细胞率最高(68.5%±31.0%);与0mg/L浓度组(26.8%±15.7%)及常规方法(38.17%±18.56%)比较,差异均有统计学意义(P<0.01和P<0.05)。[结论]苦杏仁苷有明显促进人血T淋巴细胞PCC增殖的作用;可缩短细胞分裂周期;对于辐射损伤应急生物剂量估算有重要的应用价值。[ Objective ] To explore the proliferation of premature chromosome condensation ( PCC ) in human blood T lymphoeytes induced by amygdalin, and to provide help for rapid biological dose assessment in radiation damage. [ Methods ] Peripheral blood samples were collected from 6 volunteers, and different concentrations of amygdalin were added. The best concentration for the proliferation of PPC and cell division of amygdalin was observed by the method of sister chromatid exchange ( SCE ), with the cell rate of the second generation used as its evaluating indicator. All of mitotic cycles ( G1, S, G2, M ) were observed by the common PCC ( cultured 48 h ) and the PCC used in this study ( cultured 40 h, added different concentration of amygdalin ), with the cell rate of G2+M periods as its evaluating indicator. [Results ] ①Amygdalin had a very significant effect in promoting Tlymphocytes proliferation ( P 〈 0.01-0.005 ). The best concentration of amygdalin was 200 mg/L, its cell rate of the second generation was increased by 0.74-1.60 times compared with the Omg/L group. ② Cultured with the best concentration of amygdalin, the time point of second generation cell( 5‰-9‰)was 28 h after colchieines added. It was 8 h earlier than the sample cultured without amygdalin( 4‰-9‰). ③The highest PCC cell rate in G2+M periods( 68.5%±31.0% )was observed in the medium of 200 mg/L of amygdalin plus calyculin A. Compared with the 0 mg/L group( 26.8%±15.7% )and common method group ( 38.17%±18.56% ), there were significant differences( P 〈 0.01, P 〈 0.05 ). [ Conclusion ] Amygdalin has a positive effect on the conventional chromosome of human blood T lymphocytes and PCC proliferate. It can reduce the time of cell division cycle. It is valuable for emergency biological dose assessment in radiation damage.
