摘 要:目的研究不同浓度硒、砷对体外培养的人脐静脉血管内皮细胞增殖率的影响,为硒拮抗砷的毒性作用提供理论依据。方法人脐静脉血管内皮细胞体外培养,当细胞处于对数生长期时进行实验。分为加砷组、加硒组和加砷加硒组。加砷组和加硒加砷组As^(3+)浓度分别为0、0.05、0.1、0.5、1.5、3、6、12μmol/L。加硒组硒的浓度分别为0.05、0.1、0.5、1、2μmol/L。加硒加砷组硒的浓度为0.1μmol/L。采用四唑盐(MTT)比色法测定细胞活性。结果HUVEC给予As_2O_3处理48h后,随着As^(3+)浓度的增加,对细胞生长的抑制作用逐渐增强。硒浓度为0.1μmol/L时,细胞活性高于正常对照组(P<0.05);硒浓度为1、2μmol/L时,细胞活性降低。当As^(3+)浓度为0.1μmol/L时,适量硒(0.1μmol/L)对低浓度的砷起拮抗作用(P<0.05)。结论砷可以抑制内皮细胞的生长,适量浓度的硒可以拮抗砷对细胞增殖的影响。Objectives To research the Arsenic and selenium effects on human umbilical vein vascular endothelial cells; to research the selenium effects on HUVEC injured by Arsenic; to provide a theoretical basis for the antagonistic toxic effects caused by arsenic of selenium. Metheds HUVEC were cultured in vitro. When HUVEC lied in logarithm growth, we divided them into three experiment groups which were As, Se and As+Se respectively, the ceils were treated with As2O3 at different concentrations( 0,0.05,0.1,0.5,1.5,3,6,12umol/L) for 48 h in vitro, while the group of selenium, the concentration of selenium is 0.05,0.1,0.5,1,2umol/L; In the As2O3 +Se group, the cells were treated with different concentration of As2O3 and Se(0.1umol/L) for 48h in vitro. The activities of cells were naeasured by MTT method. Results As2O3 treated HUVEC after 48 h, with the increasing concentration of As2O3, the inhibition of cell growth gradually. Cell activity was significantly higher at the dose of 0.1umol/L compared With that of control(P〈0.05), and decreased with the increased doses from 1 umol/Lto 2 umol/L. When the concentration of As3+ was 0.1umol / L, the appropriate amount of selenium took antagonism effect from arsenic (P〈0.05). Conclusion Arsenic can inhibit the growth of endothelial cells, while appropriate concentration of selenium could inhibit the effects of arsenic on the cells.
