摘 要:目的介绍一种新型疫苗递送系统"GEM微球",可用于制备诱发机体同时产生局部黏膜免疫和系统免疫的多价疫苗。方法利用乳酸菌制备GEM微球,获得融合了蛋白锚定结构(PA)的抗原或多肽等免疫原,二者结合后直接运载至黏膜作用于靶点,进而发挥诱导机体免疫应答的疫苗作用。结果与PA融合表达的蛋白,可高载量与革兰氏阳性菌递送系统"GEM微球"结合,制备多价疫苗。结论本文提供的构建"GEM微球疫苗"的相关实验技术,即GEM新型的疫苗递送系统,可为对迄今无安全有效疫苗的布鲁氏菌病的免疫防护和治疗提供有价值的依据。Objective GEM particles are suitable especially for the mucous membrane immunity vaccine, because it could induce simultaneously the partial mucous membrane immunity and the system immunity. GEM particles are one kind of new vaccine deriver system. Methods GEM particles were prepared by the food grade lactobacillus, and got fusion expressing protein which are antigens or peptides with protein anchor (PA) domain. Combining the protein and GEM particles, the complex is as the vaccine to induce immune response. Results Protein with PA can be highly loaded by grampositive enhancer matrix GEM to prepare the multivalent vaccine. Conclusion To Brucellosis and other infectious diseases, GEM particles have the extremely good application value on immunity protection and the treatment for these discases.
