摘 要:苯丙氨酸解氨酶(PAL,EC 4.3.1.5)是植物体内苯丙烷类代谢途径中的关键酶和限速酶,与植物的抗病性和植物体内黄酮类色素等多种次生物质的生物合成密切相关。利用同源克隆方法成功克隆了桑树苯丙氨酸解氨酶基因(pal)cDNA片段(GenBank登录号:FJ938356),该片段长1 105 bp,编码367个氨基酸,氨基酸序列的34-50区域为PAL的特征序列。桑树pal基因和其他植物pal基因的同源性分析表明其与甜樱桃的亲缘关系最近,同源性高达82%,与树莓的同源性达80%,与杨树和咖啡树也达到75%,由此也证实克隆的桑树cDNA片段为pal基因家族成员之一。Phenylalanine ammonia-lyase(PAL,EC 4.3.1.5) is the key and rate-limiting enzyme in plant phenylpropanoid pathway,and is considered to have a close relationship with plant resistance to diseases,biosynthesis of flavonoid pigments and other secondary substances.A cDNA fragment of pal gene was cloned successfully from mulberry(GenBank accession No.FJ938356).The cDNA was 1 105 bp long and encoded a polypeptide of 367 amino acids.Its 34th to 50th amino acids had the characteristic PAL domain.Homology analysis of mulberry pal gene with other plants revealed that it had the closest phylogenetic relationship with Prunus avium,with a sequence identity as high as 82%.Its sequence identity with Rubus idaeus was 80% and that with Populus trichocarpa and Coffea canephora was 75%,respectively.Therefore,the obtained mulberry cDNA sequence is one member of the pal gene family.
