天麻葡聚糖对CCl_4诱导的小鼠急性肝损伤的影响

摘　　要：目的探讨天麻葡聚糖对CCl4诱导的小鼠急性肝损伤的影响。方法正常小鼠随机分成正常对照组、模型组、给药组,每组10只小鼠。给药组小鼠通过灌胃方法给予天麻葡聚糖50mg/kg,连续5d;正常组和模型组小鼠通过灌胃方法给予等量0.9%NaCl溶液。末次给药24h后,模型组和给药组小鼠按0.04ml/只灌胃给予CCl4原液,造成急性肝损伤,正常组灌胃给予等量0.9%NaCl溶液;24h后测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,并进行肝组织病理学观察。结果与模型组相比,天麻葡聚糖组小鼠血清ALT活性降低极显著,AST活性变化也有降低的趋势;病理切片表明葡聚糖作用后肝组织损伤程度明显减轻。结论天麻葡聚糖对CCl4诱导的小鼠急性肝损伤具有较好的保护作用。Objective To investigate the effects of glucans from Gastrodia elata B1 on acute liver injury induced by GEl4 in mice. Methods Thirty mices were randomly divided into normal group, model group and treatment group. The former two groups were intragastrically given physiologic saline（ 50 mg/kg body weight） and the latter group was given glucans （50mg/kg body weight）, all for five days. Twenty-four hours after the last administration, model group and treatment group were given with CCl4 solution to induce acute hepatic injury, and the normal group was given physiologic saline. Twenty-four hours later, the activities of ALT and AST were detected, and the hepatic tissues were pathologically identified. Results Compared with that in the model group, the activity of ALT in the treatment group was decreased, and the degeneration and necrosis of hepatic tissues of treatment group was obviously lessened. Conclusion Glucans fror. Gastrodia clara B1. has protective effect on CCl4-induced acute liver injury in mice.
