摘  要:目的探讨牡蛎活性肽BPO G50-Ⅱ对人舌鳞癌Tca8113细胞增殖和凋亡的影响。方法不同浓度BPOG50-Ⅱ作用于体外培养的人舌鳞癌Tca8113细胞,作用24、48、72 h时计数拒染活细胞数,计算生长抑制率(IR)和IC50。采用光镜、电镜观察48 h后细胞形态及超微结构变化;荧光染色法观察BPO G50-Ⅱ作用后的凋亡情况。结果BPO G50-Ⅱ作用后IR明显降低,呈剂量与时间依赖性;24、48、72 h的IC50分别为1.50、0.57、0.20 g/L;电镜观察细胞出现明显凋亡,荧光显微镜下见细胞出现凋亡改变。结论牡蛎活性肽对人舌鳞癌细胞株Tca8113体外增殖具有抑制作用,可引起细胞形态学改变并能诱导其发生凋亡。Objective To investigate the effect of bioactive peptides from oyster ( BPO G50- Ⅱ) on the proliferation and apoptosis tongue squamous cell carcinoma Tca8113 cells in human. Methods The human tongue squamous cell carcinoma Tca8113 cells were performed with BPO G50-Ⅱ in different concentrations, the ceils of resisted-colour trypan blue were counted 24,48,72 h later, the inhibition rate (IR) and ICso were calculated. The histological and ultras- tructural changes of cell were scored using light and electron microseopy 48 h later,the apoptosis of Tca8113 cells induced by BPO G50-Ⅱ was studied by fluorescent staining. Results IR of Tca8113 ceils decreased significantly after treated by BPO G50-Ⅱ in a dosage and time dependant manner. The IC50 at 24,48 and 72 h were 1.50,0.57 and 0.20 g/L respectively. Typical apoptosis and morphology changes were foundunder the electronmicroseop and fluorescence microscope. Conclusion BPO G50-Ⅱ can inhibit the cell proliferation of human tongue squamous cell carcinoma Tca8113 cells, and cause cell morphology changes and induce apoptosis.
