摘  要:目的:探讨结缔组织生长因子(CTGF)在肾血管性高血压大鼠(RHR)动脉损害发生、发展中的作用。方法:用二肾一夹方法构建肾血管性高血压大鼠模型。32只大鼠被分为四组:假手术6周(S6)组、高血压6周(H6)组、假手术12周(S12)组、高血压12周(H12)组,采用逆转录聚合酶链式反应(RT-PCR)法检测大鼠胸主动脉CTGF、纤维连接蛋白(FN)、Ⅲ型胶原(ColⅢ)的表达水平,并用免疫组化法显示CTGF的表达。结果:高血压大鼠术后6周即出现动脉损害,FN、ColⅢmRNA表达明显上调(P<0.05),术后12周其表达进一步增加(P<0.01)。在假手术组CTGF无明显表达,而高血压组CTGFmRNA表达明显增加(P<0.01)。结论:结缔组织生长因子与肾血管性高血压大鼠动脉损害过程有重要关联。Objective: To investigate the roles of connective tissue growth factor (CTGF) on development of artery injury in renovascular hypertensive rats. Methods: Hypertension was induced by clipping the left renal artery. CTGFm- RNA, fibroneetin (FN) mRNA, collagen (Col) ⅢmRNA were detected by RT--PCR on six-week and twelve-week after operation and CTGF was also detected by immunohistochemistry. Results: The artery injury was found even on six-week. Compared with the normal control group, the rats with hypertension on 6-week and 12-week showed that the levels of CTGF mRNA, FN mRNA, Colin mRNA in thoracic aorta increased (P〈0. 01) . Conclusion: Connective tissue growth factor may significantly association with development of artery injury of renovascular hypertension.
