摘  要:目的探讨烟酰胺和四环素治疗大疱性类天疱疮(BP)的抗炎作用的可能分子机制。方法应用辛酸-硫酸胺法从BP患者疱液中纯化IgG,将纯化的4mg/mL的BPIgG与角质形成细胞共同培养24h,孵化前分别用不同浓度烟酰胺、四环素进行干预。MTT法检测各组药物对细胞增殖的影响,ELISA方法检测培养上清液中IL-8的表达。结果烟酰胺、四环素在药理浓度范围内均不影响细胞增殖。四环素以浓度依赖方式抑制BPIgG诱导角质形成细胞分泌IL-8。结论四环素可能通过抑制BPIgG诱导角质形成细胞分泌IL-8,在治疗BP中发挥抗炎作用。Objective To investigate possible molecular mechanisms of anti-inflammatory action of nicotinamide and tetracycline in the treatment of bullous pemphigoid(BP). Methods IgG in the blister fluid of BP patients was purified by caprylic acid/ammonium sulfate precipitation. The purified 4mg/ml of BP IgG and keratinocytes were co-cultured for 24h. At the beginning of this incubation,nicotinamide and tetracycline were added to the cultured medium. Cell proliferation was detected by MTT assay, the IL-8 protein level in the culture supernatant was measured by ELISA. Results In their phamacological range, nicotinamide and tetracycline did not reduce the viability of normal keratinocytes. Tetracycline inhibited the IL-8 release from cultured human keratinocytes treated with BP IgG,in a dose-dependent fashion. Conclusion Tetracycline exhibits its anti-inflammatory effect in the treatment of BP partly by inhibition of IL-8 release from cultured keratinocytes.
