摘  要:在单因素试验的基础上,用正交试验优化枣果黄酮类化合物提取的工艺条件;比较5种树脂的吸附作用,对最适树脂进行静态和动态吸附试验;进一步用硅胶柱层析纯化,薄层层析检测。最佳提取条件:60%乙醇为提取溶剂,料液比1∶20(W∶V),55℃下提取2次,每次2h,枣果黄酮类化合物的提取率可达80.78%,纯度为15.27%;HPD-417型树脂对枣果黄酮类化合物的吸附量为69.21mg/g,解吸率为90.7%。最佳操作条件:上样液浓度15mg/mL,pH6.0,流速2.0mL/min,所得枣黄酮纯度为81.59%;经硅胶柱层析进一步纯化所得枣黄酮纯度为97.98%。On the basis of single-factor test, the optimum conditions extracting flavonoids from fruit of jujube were analyzed by orthogonal test. The adsorption capabilities of five kinds of macroreticular resin were compared. Static and mobile state experiments were conducted to study adsorption capability of the most suitable resin. The crude flavonoids were purified by silica gel column chromatography, detected by thin-layer chromatography. The optimum conditions were the extraction solution of 60% ethanol, the solid-liquid ratio was 1 : 20(W : V) , at 50℃, extraction time 2 h for 2 times, the total flavonoids extraction rate is 80. 78%, purity was 15. 27%. The HPD-417 flavonoids adsorption was 69. 21 mg/g, desorption rate was 90.7%. The optimum technological parameters were the initial concentration of the flavonoids was 15 mg/mL,pH 6.0, the velocity of flow was 2.0 mL/min, purity of flavonoid was 81.59%. The purity of jujube flavonoid was 97. 98%. obtained by silica gel column chromatography.
