摘 要:目的:建立以高效液相色谱指纹图谱与多指标成分定量相结合的山楂质量评价方法。方法:采用高效液相色谱-紫外检测法,以磷酸盐缓冲液(pH 3.0)-乙腈梯度洗脱。结果:建立了山楂饮片高效液相色谱指纹图谱,指认了5个色谱峰:绿原酸,表儿茶素,原花青素B2,异槲皮素,及芦丁与金丝桃苷的共洗脱峰;测定了绿原酸与表儿茶素的含量,其中绿原酸含量为0.03%-0.06%;表儿茶素含量为0.09%-0.43%。结论:该方法可为山楂的质量评价研究提供实验数据。Objective:To establish a quality evaluation method of hawthorn using HPLC fingerprint and multi-ingredient quantification.Method:High-performance liquid chromatography coupled with a ultraviolet detector was used and the samples were performed with a mobile phase composed of phosphoric buffer(adjust pH to 3.0) and acetonitrile.Result:The HPLC fingerprint was established,and five chiromatographic peaks,such as chlorogenic acid,epicatechin,procyanidin B_2,isoquercitin,were found in all samples;the contents of chlorogenic acid and epicatechin were in the range of 0.03% to 0.06% and 0.09% to 0.43%,respectively.Conclusion:The method can provide some useful experimental data for quality evaluation of hawthorn.
