摘 要:目的:观察消渴通络颗粒对糖尿病大鼠周围神经病变的影响,并初步探讨其作用机制。方法:采用链脲佐菌素(STZ)诱发糖尿病大鼠模型,成模大鼠分别用高、中、低剂量的消渴通络颗粒连续灌胃给药8周,观察药物对糖尿病大鼠空腹血糖、糖化血红蛋白、坐骨神经中醛糖还原酶(AR)和Na+-K+-ATP酶活性以及组织形态学的影响,并在实验的第4周和第8周末分别监测大鼠坐骨神经运动传导速度(MNCV)及热痛阈。结果:与模型组相比,消渴通络颗粒能明显降低糖尿病大鼠坐骨神经AR活性,升高Na+-K+-ATP酶活性。对糖尿病大鼠坐骨神经传导速度、热痛阈有明显改善作用,也可减轻坐骨神经髓鞘水肿,改善神经病变。结论:消渴通络颗粒对糖尿病大鼠周围神经病变有明显的改善作用,其作用可能与其改善糖尿病大鼠代谢紊乱、抑制多元醇通路等有关。Objective:To observe the effects of X iaoketongluo granula (XKTL) on the diabetic peripheral neuropathy ( DPN ) in rats, and to investigate the possible mechanism. Methods : Diabetes was induced by streptozocin(STZ). Diabetic rats were treated with oral XKTL at 3 doses. After 8-week XKTL treatment, the effects on the FBG, HbA 1 c, aldehyde reduetase ( AR ) , Na^+ -K^+ -ATPase and morphological changes were observed. The conduction velocity of sciatic nerve and the pain threshold detected by illumination test were observed, respectively, at the end of the 4th and 8th week. Results:As compared with diabetic control,XKTL reduced AR activity and increased Na^+ -K ^+-ATPase activity in sciatic nerves of diabetic rats. Meanwhile, it obviously improved the nerve conduction velocity and the pain threshold, and alleviated the pathological changes ( myelin sheathe swollen of the sciatic nerves) in diabetic rats. Conclusion:XKTL alleviates the lesion of DPN in rats; this may relate to the improvement of metabolic disturbance and the inhibition of the polyol pathway.
