摘  要:以龙眼"红核子"品种为材料,研究甘露醇与肌醇交互培养及蔗糖含量、琼脂含量、盐浓度、PP333浓度、光质等对龙眼胚性愈伤组织(Embryogenic callus,EC)限制生长保存的影响,结果表明:同时添加20g/L甘露醇、0.10g/L肌醇,并把接种量控制在0.1g/瓶,龙眼EC的继代周期可进一步延长到70d左右。同时,以"红核子"、"松风本"和"聚龙105"为材料,对保存的龙眼EC及其体胚发生过程中染色体数目的变异进行研究,结果表明:染色体变异始于胚性愈伤组织,但有趋于稳定的趋势。‘Honghezi' Longan(Dimocarpus longan ‘Honghezi') was used to study the effects of mannitol and inositol cross-culture, sucrose, agar, salt, PP333 and light quality on the minimal growth of longan embryogenic callus(EC) for conservation. The results showed that the combined addition of 20 g/L mannitol and 0.1 g/L inositol with the inoculation amount of 0.1 g per bottle, prolonged the, subculture cycle to more than 70 days. At the same time, the long cuhivars ‘Honghezi', ‘Songfengben' and ‘Julong 105' were used to investigate the variation of chromosome number of longan embryogenic callus during somatic embryogenesis. The results indicated that the chromosome variation occurred at the embryogenic cell stage and then tended to be stable.
