摘 要:目的研究海带褐藻多糖硫酸酯(fucoidan from Laminaria japonica,FL)对四氯化碳所致小鼠急性肝损伤的保护作用。方法小鼠50只,随机分为正常组、模型组、阳性药对照组以及高、低剂量给药组,阳性药对照组灌胃联苯双酯100mg·kg^(-1),给药组分别按200,100mg·kg^(-1)剂量灌胃褐藻多糖硫酸酯,每天1次,连续给药10d。末次给药后1h,除正常对照组外其余各组均腹腔注射10%四氯化碳溶液0.1mL·(10g)^(-1)。24h后,取血测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)和总胆红素(BIL)水平,取肝脏称重,计算肝脏指数,同时观察肝脏病理学变化。结果高剂量的褐藻多糖硫酸酯能显著降低肝损伤小鼠血清ALT、AST和BIL水平,肝脏指数明显下降,病理观察结果也显示能减轻小鼠的肝损伤程度。结论褐藻多糖硫酸酯对四氯化碳所致小鼠急性肝损伤有保护作用。Objective To study the protective effects of fucoidan from Laminaria japonica on CCl4-induced acute liver injury in mice. Methods Fifty mice were allocated into control group, model group, bifendate group, low and high dosage of fucoidan group randomly. The mice were treat with hifendate(100mg · kg^-1) or fucoidan (200,100mg · kg^-1) for 10 days. Then 10%CCl4 (10mL · kg^-1) was given to mice by intraperitoneal injection. The activities of serum ALT,AST,ALP and BIL were determined, the body weight and liver weight were measured, as well as the pathological changes of the liver were observed in all groups. Results fucoidan (200mg · kg^-1) could significantly lower the ALT,AST BIL in serum and the liver index of mice, improve the liver histology. Conelution fucoidan can protect the liver against damage induced by CCl4 in mice.
