摘 要:目的:探讨三氯乙烯(TCE)染毒L02细胞后Foxp3和GATA3基因mRNA表达水平改变。方法:培养L02细胞,用TCE进行染毒,3个实验组TCE剂量分别是0.25、0.5和1.0 mmol/L,染毒时间12 h,另设1个对照组不染毒TCE。染毒结束后提取细胞总RNA,用实时荧光定量PCR测定Foxp3和GATA3的mRNA表达水平。结果:低剂量TCE0.25 mmol/L组Foxp3 mRNA表达水平与对照组比较升高(P<0.05),中剂量和高剂量组(TCE0.5和1.0 mmol/L)Foxp3mRNA表达水平与对照组比较显著升高(P<0.01);同样,TCE 0.25、0.5和1.0 mmol/L实验组GATA3 mRNA表达水平与对照组比较都明显升高,差异有统计学意义(P<0.05或P<0.01)。结论:三氯乙烯染毒L02细胞后可引起免疫相关基因Foxp3和GATA3 mRNA表达水平升高,提示三氯乙烯导致机体细胞改变可能与免疫基因有关。Objective:To Study mRNA expression of Foxp3 and GATA3 genes in L02 cells after treatment with trichloroethylene(TCE).Methods:L02 cells were treated with trichloroethylene at doses of 0.25,0.5 and 1.0 mmol/L for 12 h,the control was not treated with TCE.After treatment the total RNA of cells was extracted,and real-time fluorescent PCR was applied to detect mRNA expression of Foxp3 and GATA3 genes in L02 cells after treatment with trichloroethylene.Results:mRNA expression of Foxp3 in three groups treated with TCE was higher than that in control(P0.05,or P0.01),additionally,TCE also induced higher levels of GATA3 mRNA expression in the groups treated with 0.25,0.5 and 1.0 mmol/L in comparison with control.Conclusion:Trichloroethylene could bring about mRNA expression alteration of Foxp3 and GATA3 genes,which indicates that the adverse effects of trichloroethylene might be associated with immunological genes.
