灵芪胶囊对肺癌A549细胞的VEGF、MMP-2、MMP-9表达的影响

摘　　要：目的:研究灵芪胶囊对肺癌A549血管内皮生长因子(VEGF),基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达的影响,以探讨其抗肿瘤作用机制。方法:本实验采用血清药理学方法制备含药血清,分别采用RT-PCR法和免疫组织化学方法检测含药血清对A549细胞VEGF mRNA和MMP-2、MMP-9蛋白表达的影响。结果:高、中、低各给药组VEGF mRNA校正值(VEGF/β-actin),分别为(0.804±0.020),(0.836±0.022),(0.914±0.009),空白对照组为(0.905±0.033),阳性对照组为(0.766±0.014)。各给药组表现出明显剂量依赖性且与空白对照组比较差异有显著性意义(P<0.01)。图像分析技术处理结果显示给药组MMP-2,MMP-9蛋白低表达(P<0.05)。结论:灵芪胶囊通过抑制肺腺癌A549VEGF mRNA、MMP-2和MMP-9的表达,从而抑制肿瘤细胞增殖生长及恶性化转移。Objective：In occurrence and development process oflung cancer, the only known angiogenesis is one element ofthe growth, proliferation and aversion oftumor cells.VEGF, MMP-2 and MMP-9 play important role in the development and aversion ofA549 cell.This experiment is to reveal the effect ofLingqi Capsule on expression VEGF, MMP-2 and MMP-9 in A549 cell and mechanism ofanti-tumor.Methods：Medicated serum was prepared with serum-pharmacology methods, the the effect ofmedicated serum on VEGFmRNA and protein expressions ofMMP-2 and MMP-9 was detected by RT-PCR technology and immunohistochemistry respectively.Results：It suggested that the VEGF mRNAcorrect value ofthe administration group was respectively（0.804±0.020）,（0.836±0.022）and（0.914±0.009）,the blank control group was（0.905± 0.033）and positive control group was（0.766±0.014）.Difference between the administration group and the blank control group was significant（P0.05）.The result with image analysis showed the low expression ofMMP-2, MMP-9 in the administration group（P0.05）.Conclusion：Lingqi Capsule inhibited the growth and malignant transformation oftumor cell by inhibiting the proliferation ofvascular endothelial cells and the expression ofMMP-2, MMP-9 in A549 cells.
