摘 要:利用随机微卫星扩增多态性DNA(RMAPD)分子标记技术,对全国范围内的12个天然居群245个个体进行遗传多样性分析,用筛选出的18对引物组合共扩增出303个条带,其中多态性条带229个,多态条带百分率为77.58%。用NTSYS-PC软件中的SIMQUAL程序计算Jaccard遗传相似性系数(genetic similarity,SG)并聚类。结果表明:245份油松材料间的SG值变化范围为0.5238~0.9682,平均为0.8141。采用UPGMA法在0.68水平上可将供试材料聚为4类。内蒙古宁城油松、山西地区油松各自聚为一类外,甘肃小陇山地区油松分别于陕西的洋县、留坝、周至、洛南、柞水的油松聚为一类,黄龙、黄陵、府谷的油松聚为一类。表明各居群间存在较高遗传多样性。Random Microsatellite Amplified Polymorphic DNA (RMAPD) markers were used to measure genetic diversity and genetic structure of 245 individuals from 12 natural populations of Pinus tabulaeformis Cart. A total of 303 loci were identified from 18 RMAPD primer combinations, amongwhich 229 loci were polymorphic and accounted for 77.58 percent. Jaccard's genetic similarity coefficients were calculated and a dendrogram was generated by unweighted pair group method of arithmetic average using NTSYS-PC. Result showed that the genetic similarity coefficients ranged from 0.523 8 to 0. 968 2 with an average of 0. 814 1. All the 245 materials were classified into 4 groups at level 0.68. Materials in Shanxi Province and those in Inner Mongoha were separately clustered into one group; meanwhile, materials in Gansu Province were clustered into one group with those from Yangxian, Liuba, Zhouzhi, Lounan, Zhashui of Shaanxi Province, and materials in Fugu were clustered into one group with those from Huanglong and Huangling of Shaanxi Province. It is proved that higher diversities exist among the populations of P. tabulaeformis in the main distribution areas in China.
