帕屈磷酸钠对W_(256)肿瘤引起大鼠骨溶解作用的影响

摘　　要：目的研究帕屈磷酸钠对Ｗ２５６肿瘤引起大鼠骨溶解作用的影响。方法除正常组以外，每鼠右后肢胫骨上端骨旁肌肉内注入０．２ｍｌＷ２５６瘤细胞悬液（４×１０６个瘤细胞），ｄ１２可引起大鼠局部骨组织溶解、破坏及血钙和钙排泄增加等变化。结果帕屈磷酸钠（ＡＰＤ，７．５、１５与３０ｍｇ·ｋｇ－１），一次性尾静脉注射对Ｗ２５６性溶骨大鼠的血钙和尿钙升高以及骨矿含量的降低均有明显的防治作用，经ＰＯＳ型显微光密度仪对鼠股骨和胫骨Ｘ光片扫描分析的结果表明，ＡＰＤ（３０ｍｇ·ｋｇ－１）对Ｗ２５６引起的大鼠骨密度降低也有明显的防治作用。结论帕屈磷酸钠对Ｗ２５６引起的大鼠溶骨及并发症具有良好的防治作用。AIM Effects of aminopropanediphoosph onate pamidronete (APD) on W 256 tumor-induced bone destruction were studied in rats.METHODS The intramusculer injection of W 256 tumor cells (0 2 ml,4×10 6 cells) beside the tibia of rats after 12 days induced osteolytic metasteses and bone destruction. RESULT APD(7 5、15 and 30 mg·kg -1 ) significinently inhibited and prevented the rised calcmia and overexcretion of calciuia of the W 256 rats. The results also showed that APD (30 mg·kg -1 )prevented and increased the density of diaphysis of the W 256 rats determined by Xra as well as microradiographically upon termination of the experiment. CONCLUSION APD has a effect preventing the high calcemia and overexcretion of calciuia and low density of diaphysis caused by W 256 tumor.
