摘  要:目的观察脑出血大鼠神经营养因子-3(NT-3)的表达情况,以及脑溢安对NT-3表达的影响,探讨脑溢安治疗脑出血的可能机制。方法80只大鼠随机分为正常组、假手术组、模型组及脑溢安组,用Ⅶ型胶原酶立体定位注射于苍白球建立脑出血模型,术后1、4、7、14、21d分别用免疫组织化学及原位杂交方法观察NT-3蛋白和mRNA的表达。结果正常组和假手术组大鼠脑内未见NT-3蛋白表达,正常组、假手术组大鼠脑内皮质和海马均可见NT-3 mRNA表达,正常组与假手术组相比较,差异无统计学意义(P>0.05);模型组和脑溢安组大鼠血肿周围、海马和脑内皮质自造模1d后,可见NT-3蛋白和mRNA表达,4d达到高峰,7d开始下降;与模型组相比较,脑溢安组大鼠脑内上述区域NT-3蛋白和mRNA在各个时间点的表达均显著增加(均P<0.01)。结论脑出血后大鼠脑内有NT-3的表达,脑溢安上调NT-3的表达,这可能是脑溢安防治脑出血的机制之一。Objective To observe the expression of neurotrophin 3(NT-3) in rats brain of experimental intraeerebral hemorrhage(ICH). To discuss the mechanism of Naoyian(NYA) to treat ICH. Methods Eighty rats were randomly divided into four groups including controlgroup(n=5) ,sham operationgroup(n=25),modelgroup(n=25) andNYA group(n=25). Theintracere bral hemorrhage of rats was induced by injecting 0.4u Ⅷ collagenase into right globus pallidus by stereotaxic apparatus. The expression of NT-3 protein and mRNA was observed by immunohistoehemistry staining and in situ hybrid technique at 1,4,7,14,21d after operation. Results NT-3 protein did not express in control group and sham operation group. NT-3 mRNA was observed to express in cortices and hippocampus of the rats in control group and sham operation group, there was no significant difference be tween the two groups (P〉0.05). NT 3 protein and mRNA were observed to express in the marginal zones of the hematoma,hippocampus and cortices of the rats in model group and NYA group after ld of ICH injury,they reached the peak at 4d,then de creased at 7d. The expression of NT-3 protein and mRNA in NYA group was more intensive than model group (P〈0.01). Conclusion NT-3 expresses in the brains of intracerebral hemorrhagic rats,NYA could promote the expression level of NT-3 protein and mRNA. It may be one of the protecting mechanisms of NYA against ICH.
