摘  要:目的探讨维生素D受体(VDR)基因FokI的多态性与T2DM及其合并动脉粥样硬化(AS)之间的相关性。方法通过提取全血DNA,PCR扩增VDR基因,FokI酶切扩增产物,检测大连地区T2DM合并AS者(AS组),DM非AS者(non-AS组)及健康对照者(NC组)的基因型。应用单因素非条件logistic回归分析研究VDR基因变异对T2DM及合并AS的影响。结果 (1)VDR基因FokI酶切位点各基因型在T2DM组和NC组中分布具有统计学差异(P<0.01)。(2)AS组和non-AS组VDR基因FokI酶切各基因型分布无统计学差异(P>0.05)。(3)VDR等位基因f与T2DM呈显著正相关,等位基因F与T2DM呈显著负相关。结论在大连地区的汉族人群中,VDR基因FokI的多态性与T2DM相关,而与T2DM合并AS无关。等位基因f是T2DM的易感基因,而等位基因F对T2DM具有保护作用。Objective To investigate the relationship of vitamin D receptor (VDR)gene FokⅠ polymorphisms with T2DM and the combined T2DM and atherosclerosis (AS). Methodds VDR gene polymorphism analysis was performed with polymerase chain reaction-restriction fragment length polymorphism in three groups. The genotype frequencies of VDR were calculated according to Hardy-Weinberg equilibrium formula. Using single factor nonconditional logistic regression analysis, the incidence of VDR gene mutation was studied. Results (1) There was significant difference in the frequency distribution of VDR Fokl genotype hetween T2DM and normal eontrols(P〈0.01). (2)There was no significant difference between T2DM and the combined T2DM and AS in VDR FokⅠ genotypes(P〉0.05). (3)Allele f was positively correlated to T2DM, whereas allele F was negatively correlated to T2DM. Conclusions VDR FokⅠ polymorphisrns is related to T2DM,but not to the combined T2DM and AS. Allele f may be a susceptible gene contributing to the development of T2DM, and allele F may be a protective gene for T2DM.
