摘  要:目的:通过检测不同比例饮食限制对运动大鼠脑组织5-HT含量及其关键酶色氨酸羟化酶的影响,探讨饮食限制与运动性中枢疲劳的关系;方法:以40只成年雄性SD大鼠为实验对象,建立不同限制饮食动物模型。实验分组为安静对照组(NC)、100%饮食运动组(100%DR)、70%饮食运动组(70%DR)、50%饮食运动组(50%DR)、30%饮食运动组(30%DR),每组8只,饮食限制时间为10天,运动方案为每天进行游泳训练30min。采用高效液相色谱法测定脑干区5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)含量;采用RT-PCR分析脑干色氨酸羟化酶TPHmRNA表达量的变化;结果:1)各运动组大鼠脑干5-HT含量均高于安静对照组,但仅有30%饮食运动组有显著性增加(P<0.05);2)5-羟吲哚乙酸含量表现为30%DR组显著高于对照组和100%DR组(P<0.05);3)大鼠脑干区TPHmRNA的表达水平显示,30%DR组显著高于NC组和100%DR运动组(P<0.05),与100%DR组相比,各限食组均有显著性差异(P<0.05);结论:不同比例的饮食限制导致大鼠脑干5-HT和5-HIAA的生成增加,且增加量与饮食限制程度有关,其原因可能与饮食限制促进脑干TPH-mRNA的表达有关。Objective:To investigate the relationship between the central fatigue and dietary restriction and examine the contents of 5-HT and 5-HIAA and its key enzyme TPH in rat brain stem respond to dietary restriction. Methods:40 male Sprague-Dawley rats were used to establish different levels of dietary restricted models (normal control group, 100% dietary restriction group, 70 % dietary restriction group, 50 % dietary restriction group, 30 % dietary restricted group, dietary restriction duration of 10 days). Concentrations of 5-hydroxy-tryptamine (5- HT), 5-hydrox-yindolacetic acid (5-HIAA) in brain stem were measured using High Performance Liquid Chromatography (HPLC), and the expression of TPH mRNA in brain stem was determined by semi-quantitative reverse-transcription polymerase chain reaction (RT-PCR). Results.. 1. Compared with the normal control group, the concentrations of 5-HT in the brain stem increased significantly in 30 %dietary restricted group (P〈0.05). 2. Compared with the normal control group and 100 % dietary restricted group, the concentrations of 5-HIAA in the brain stem increased significantly in 30%dietary restricted group (P〈0.05). 3. The expressions of TPH mRNA in the brain stem shows that compared with the normal control group and 100 % dietary restricted group, the expressions of TPH mRNA in the brain stem have been enhanced significantly in 30 % dietary restricted group ( P〈0.05 ). Compared with 100 % dietary restriction group, all of the dietary restriction groups have been enhanced significantly (P 〈0.05). Conclusions:Dietary restriction enhances the metabolism of 5-HT in the brain stem of rats, which goes along with the different levels of dietary restricted.
