摘  要:目的:探讨白藜芦醇(Res)对小鼠胚胎成纤维细胞(MEF)增殖和细胞周期的影响及变化规律。方法:通过不同浓度(1~150μmol/L)Res作用于增殖期的MEF,用MTT法检测72h后各组细胞增殖变化,用流式细胞术检测并分析其细胞周期。结果:Res1μmo/L组A值明显高于对照组(P<0.05),Res50μmol/L和150μmol/L组A值分别为0.196±0.055和0.185±0.034,均明显低于对照组(分别P<0.05,P<0.01),其细胞生长抑制率分别为14.0%和18.9%。细胞周期检测结果表明,Res1μmol/L组中G2/M期细胞占(14.82±0.49)%显著高于对照组(P<0.01)。50μmol/L和150μmol/L组G0/G1期细胞所占比例分别为(59.61±6.03)%和(65.31±0.27)%均显著高于对照组(P<0.01)。结论:Res对MEF增殖的影响,具有双向浓度依赖性,增殖和抑制增殖均与细胞周期调控有关,其中阻断G1期细胞进入S期是其抑制MEF增殖主要因素。AIM: To discuss the cell multiplication and cell cycle situation in mouse embryonic fibroblast ( MEF), which acted by Resveratrol (Res). METHODS: Res with different concentration (0. 15 - 500 μmol/L) was used on MEF. After 72 h, cell multiplication situation and Inhibitory rate of cell growth were tested by MTT, and cell cycle was analyzed by using flow cytometry. RESULTS: A of the group with 1μmol/L Res was Significant higher than the control group ( P 〈 0.05 ). A of groups with 50 μmol/L and 150 μmol/L were 0.196 ±0.055 and 0. 185 ± 0.034 separately, which were lower than the control group ( P 〈 0.05, P 〈 0.01 separately), and the inhibitory rates of cell growth were 14. 0% and 18. 9% separately. Cell cycle analysis showed that the group with 1 μmol/L Res had ( 14.82 ± 0.49)% cells in G2/M period, which was higher than the control group ( P 〈 0.01 ). The 50μmol/L and 150 μmol/L groups had (59.61±6.03)% and (65.31±0.27)% cells in G0/G1 period, those were higher than the control group (P〈0.01). CONCLUSION: Two-way effect, which relies on the dose of Res, is showed on MEF. Multiplication and growth inhibition had relation with the control of cell cycle. Prevent cell turn into S period from G1 period in cell cycle is main inhibition growth effect in MEF.
