摘 要:目的:研究抗纤灵冲剂对单侧输尿管梗阻(UUO)大鼠肾组织氧化应激的影响。方法:SD雄性大鼠120只,随机分为正常组、假手术组、模型组、抗纤灵组和科素亚组,各组分别观察28d。于造模后每周各组随机选取6只检测肾组织匀浆超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)。结果:与正常组、假手术比较,模型组肾组织SOD、GSH明显降低(P<0.01),MDA明显升高(P<0.01)。在UUO模型中,SOD、GSH、逐渐下降,MDA逐渐升高。抗纤灵组与模型组比较,肾组织SOD、GSH明显升高(P<0.01),MDA明显下降(P<0.01)。结论:在肾间质纤维化的起始和发展过程中,都有氧化应激的参与。抗纤灵冲剂能有效缓解在大鼠UUO模型中肾组织氧化应激的反应,进而可能延缓肾间质纤维化的进程。Objective:To explore effect of Kang xian ling decoction (KXL) on oxidation stress reaction in rat kidney after unilateral urethral obstruction.Methods:One hundred and twenty male Sprague-Dawley rats were randomly divided into 5 groups:the normal group,the sham-operation group,the model group,the KXL group,and the Lostartan group.Each group was randomly divided again into subgroups (each n=6) according to the times (7 d,14 d,21 d,28 d).Malondialdehyde (MDA),superoxide dismutase (SOD),glutathione (GSH) contents in obstructed kidney tissue of rats were tested.Results:Compared with those in the normal group and the sham-operation group,SOD and GSH contents in the model group were decreased (P0.01),while the content of MDA increased (P0.01).Except for the normal group and the sham-operation group,the contents of SOD and GSH in other groups were decreased gradually,while the content of MDA was increased gradually.Compared with those in the Model group,SOD and GSH contents in the KXL group increased (P0.01),while the content of MDA decreased (P0.01).Conclusion:Oxidation stress reaction exists in the onset and development of renal interstitial fibrosis.Kang xian ling decoction may degrade renal interstitial fibrosis by suppressing oxidative stress reaction in rats after UUO.
