丹参注射液对腹膜纤维化大鼠腹膜通透性的影响

摘　　要：目的:探讨中药丹参对大鼠腹膜透析模型腹膜形态及功能的影响。方法:将56只雄性SD大鼠随机分为正常组、模型组、丹参低剂量组、丹参中剂量组、丹参高剂量组。以腹腔注射脂多糖复制动物模型,并予相应药物干预,实验周期28d。结果:丹参组在2h腹透液中Scr、BUN、Glu水平、网膜透光度、抑制血管增生、降低腹透液NO含量方面优于模型组,在腹透液超滤、2h腹透液中白蛋白水平、腹透液中K+水平与模型组差异无统计学意义。病理上丹参组在改善炎症、血管扩张出血方面并无明显优势,但能延缓纤维组织增生,光镜、电镜显示丹参可延缓腹膜纤维化的程度,以中剂量丹参治疗组作用最明显。结论:加入脂多糖的4.25%葡萄糖腹透液可损伤腹膜结构和功能,丹参能够拮抗脂多糖对腹膜间皮层结构的损伤,减少基质蓄积,维持了腹膜间皮层结构的完整,改善腹膜结构,提高透析效能,延缓腹膜纤维化。Objective：To explore the effect of traditional Chinese medicine Salviae on peritoneal morphology and function of rat model of peritoneal dialysis.Methods：Fifty-six male SD rats were randomly divided into the control group,the model group,the low-dose of Salvia group,the middle dose of Salvia group,the high dose of Salvia group.Results：In respect of the levels of Cr、BUN、Glu,light transmission omentum in 2 h peritoneal dialysis fluid,angiogenesis inhibition,lower NO concentrations in peritoneal dialysis fluid,the Salviae group was better than the model group but there was no difference with the model group in respect of the ultrafiltration in peritoneal dialysis fluid,albumin levels in 2 h peritoneal dialysis fluid,and K＋ level in peritoneal dialysis fluid.There was no obvious advantage in respect of vascular ectasia or bleeding in the SM group,compared with other groups.But Salviae can delay the proliferation of fibrous tissue in peritoneum,which was proved by light microscopy and electron microscopy,especially in the middle dose of Salviae group.Conclusion：Lipopolysaccharide added to 4.25% glucose peritoneal dialysis fluid can damage the structure and function of peritonium,and salviae can be antagonistic about this effect through reducing matrix accumulation and maintaining the integrityand and the structures of the cortex,It improves the effectiveness of dialysis and delays peritoneal fibrosis.
